Formula for Oligo Dilution
This tool uses the following formula to determine the required volume of diluent needed to achieve a desired oligonucleotide concentration:
Required volume (V1) = desired final volume (V2) × desired concentration (M2) / stock concentration (M1)
M1 × V1 = M2 × V2 represents the principle of conservation of mass in the context of dilution, ensuring the moles of solute before and after dilution remain constant.
Applications of the Oligo Dilution Calculator
- Primer Preparation for PCR: The accuracy of primer concentrations directly affects the efficiency and specificity of PCR assays. Our calculator helps in precisely diluting stock primers to optimal concentrations necessary for robust PCR performance.
- Synthetic Gene Assembly: When constructing synthetic genes, multiple oligonucleotides must be assembled at precise molar ratios. The calculator assists in achieving these ratios by accurate dilution, crucial for successful gene synthesis.
- Quantitative PCR Setup: For qPCR, precise primer and probe concentrations are critical for quantification accuracy. The calculator ensures that these components are diluted to the exact concentrations needed for consistent and reproducible results.
- Labeling Reactions: Diluting oligonucleotides to specific concentrations is essential for efficient labeling with fluorescent dyes or radioactive isotopes used in various detection and imaging techniques.
