  • Design LNA-enhanced primers: Use Locked Nucleic Acid (LNA)-enhanced primers to increase binding affinity and specificity – effectively discriminating between closely related miRNAs and lower off-target amplification
  • Adjust primer Tm: Try a primer melting temperature of about 59°C to improve binding to short miRNA sequences
  • Control your assay: Validate assay specificity by testing sequences with single nucleotide mismatches and with no template controls to assure no cross-reactivity or false positives; include spike-in RNAs to monitor extraction/RT efficiency and positive controls for target detection
