How to Set Up PyroMark CpG Assays in PyroMark Q48 Software

This tutorial shows you how to manually set up PyroMark CpG assays in the PyroMark Q48 software using assay information provided on GeneGlobe. You will learn how to locate the required sequence data, transfer it into the software, and generate a correct assay setup for accurate CpG methylation analysis.

Step-by-step guidance on manually setting up PyroMark CpG assays in the PyroMark Q48 software using sequence data from GeneGlobe.

Understanding PyroMark Q48 CpG Assay setup

Unlike PyroMark Q24, Q24 Advanced, or Q96 instruments — where assay setup files can be downloaded directly — PyroMark Q48 assays must be configured manually. The required Sequence to analyze is available on the GeneGlobe product page and must be copied into the PyroMark Q48 software during assay creation. This tutorial explains where to find this information and how to use it correctly to ensure reliable and reproducible CpG methylation results.

What you will learn in this tutorial

In this tutorial, you will learn how to:

  • Locate PyroMark CpG assay resources on GeneGlobe
  • Identify and copy the correct Sequence to Analyze
  • Create a new CpG assay in the PyroMark Q48 software
  • Automatically generate the correct dispensation order
  • Save the assay setup file for future use

Step-by-step workflow for PyroMark Q48 Assay setup

Find the Assay Information on GeneGlobe

Navigate to the PyroMark CpG assay product page on GeneGlobe and open the Product Specifications section.

Review Available Product Resources

  • Download setup files if you are using PyroMark Q24 or Q96 instruments.
  • For PyroMark Q48 assays, use the displayed graph and sequence section to locate the Sequence to Analyze.

Copy the Sequence to Analyze

Scroll to the bottom of the product page, highlight the Sequence to Analyze, and copy it to your clipboard.

Create a New CpG Assay in PyroMark Q48 Software

  • Open the PyroMark Q48 software.
  • Go to: File → New Assay → CpG Assay.
  • Paste the copied sequence into the field: Sequence Before Bisulfite Treatment.

Generate the Dispensation Order

Use the software’s built-in function to automatically calculate the dispensation order based on the entered sequence.

Save the Assay Setup

Save the completed assay file for future use. This ensures a repeatable and consistent workflow for your experiments.

Who This Tutorial Is For
